    The role of the Remotely Augmented Vehicle (RAV) laboratory in flight research

    An overview is presented of the unique capabilities and historical significance of the Remotely Augmented Vehicle (RAV) Lab at NASA-Dryden. The role is reviewed of the RAV Lab in enhancing flight test programs and efficient testing of new aircraft control laws. The history of the RAV Lab is discussed with a sample of its application using the X-29 aircraft. The RAV Lab allows for closed or open loop augmentation of the research aircraft while in flight using ground based, high performance real time computers. Telemetry systems transfer sensor and control data between the ground and the aircraft. The RAV capability provides for enhanced computational power, improved flight data quality, and alternate methods for the testing of control system concepts. The Lab is easily reconfigured to reflect changes within a flight program and can be adapted to new flight programs

    Growth, water use efficiency and stable carbon isotopes in commercial clones of Eucalyptus

    The expansion of Eucalyptus plantations to supply timber for an increasing population in South Africa will result in a great reduction in the country's run-off water. If Eucalyptus continues to be the source of timber in South Africa, the selection of more water use efficient species for planting in existing and new areas has to be implemented. An understanding of the physiological factors ruling growth and water use efficiency in Eucalyptus is needed to develop selection criteria for improved water use efficiency and harvestable stem production under a limited water supply. This study investigated the effects of soil moisture availability on the growth and water use efficiency of 6 commercial clones of Eucalyptus commonly grown in South Africa with the aim of determining the following: (i) The extent of clonal variation in growth, dry mass allocation patterns, water use efficiency and the water cost of wood production at 16 months after planting. (ii) The influence of plant physiological traits such as patterns of dry mass allocation, canopy leaf area, leaf canopy density, specific leaf area, foliar nitrogen concentration and instantaneous rates of photosynthesis and transpiration, on growth, water use efficiency and the water cost of wood production. (iii) The complications associated with sampling for stable carbon isotope ratios (δ¹³C) within a eucalypt canopy and the potential use of δ¹³C in plant tissues as a tool for ranking clonal water use efficiencies

    Experience with Ada on the F-18 High Alpha Research Vehicle Flight Test Program

    Considerable experience was acquired with Ada at the NASA Dryden Flight Research Facility during the on-going High Alpha Technology Program. In this program, an F-18 aircraft was highly modified by the addition of thrust-vectoring vanes to the airframe. In addition, substantial alteration was made in the original quadruplex flight control system. The result is the High Alpha Research Vehicle. An additional research flight control computer was incorporated in each of the four channels. Software for the research flight control computer was written in Ada. To date, six releases of this software have been flown. This paper provides a detailed description of the modifications to the research flight control system. Efficient ground-testing of the software was accomplished by using simulations that used the Ada for portions of their software. These simulations are also described. Modifying and transferring the Ada for flight software to the software simulation configuration has allowed evaluation of this language. This paper also discusses such significant issues in using Ada as portability, modifiability, and testability as well as documentation requirements

    A simulation evaluation of a four-engine jet transport using engine thrust modulation for flightpath control

    The use of throttle control laws to provide adequate flying qualities for flight path control in the event of a total loss of conventional flight control surface use was evaluated. The results are based on a simulation evaluation by transport research pilots of a B-720 transport with visual display. Throttle augmentation control laws can provide flight path control capable of landing a transport-type aircraft with up to moderate levels of turbulence. The throttle augmentation mode dramatically improves the pilots' ability to control flight path for the approach and landing flight condition using only throttle modulation. For light turbulence, the average Cooper-Harper pilot rating improved from unacceptable to acceptable (a pilot rating improvement of 4.5) in going from manual to augmented control. The low frequency response characteristics of the engines require a considerably different piloting technique. The various techniques used by the pilot resulted in considerable scatter in data. Many pilots readily adapted to a good piloting technique while some had difficulty. A new viable approach is shown to provide independent means of redundancy of transport aircraft flight path control

    Variation in general practitioners' information-seeking behaviour:a cross-sectional study on the influence of gender, age and practice form

    Objective: To assess general practitioners’ (GPs’) information-seeking behaviour and perceived importance of sources of scientific medical information and to investigate associations with GP characteristics. Design: A national cross-sectional survey was distributed electronically in December 2013. Setting: Danish general practice. Subjects: A population of 3440 GPs (corresponding to approximately 96% of all Danish GPs). Main outcome measures: GPs’ use and perceived importance of information sources. Multilevel mixed-effects logit models were used to investigate associations with GP characteristics after adjusting for relevant covariates. Results: A total of 1580 GPs (46.4%) responded to the questionnaire. GPs’ information-seeking behaviour is associated with gender, age and practice form. Single-handed GPs use their colleagues as an information source significantly less than GPs working in partnership practices and they do not use other sources more frequently. Compared with their younger colleagues, GPs aged above 44 years are less likely to seek information from colleagues, guidelines and websites, but more likely to seek information from medical journals. Male and female GPs seek information equally frequently. However, whereas male GPs are more likely than female GPs to find that pharmaceutical sales representative and non-refundable CME meetings are important, they are less likely to find that colleagues, refundable CME meetings, guidelines and websites are important. Conclusion: Results from this study indicate that GP characteristics should be taken into consideration when disseminating scientific medical information, to ensure that patients receive medically updated, high-quality care.KEY POINTS Research indicates that information-seeking behaviour is associated with GP characteristics. Further insights could provide opportunities for targeting information dissemination strategies. Single-handed GPs seek information from colleagues less frequently than GPs in partnerships and do not use other sources more frequently. GPs aged above 44 years do not seek information as frequently as their younger colleagues and prefer other information sources. Male and female GPs seek information equally frequently, but do not consider information sources equally important in keeping medically updated

    Tuberculosis among economic migrants: a cross-sectional study of the risk of poor treatment outcomes and impact of a treatment adherence intervention among temporary residents in an urban district in Ho Chi Minh City, Viet Nam.

    BACKGROUND Tuberculosis (TB) remains a major cause of avoidable deaths. Economic migrants represent a vulnerable population due to their exposure to medical and social risk factors. These factors expose them to higher risks for TB incidence and poor treatment outcomes. METHODS This cross-sectional study evaluated WHO-defined TB treatment outcomes among economic migrants in an urban district of Ho Chi Minh City, Viet Nam. We measured the association of a patient's government-defined residency status with treatment success and loss to follow-up categories at baseline and performed a comparative interrupted time series (ITS) analysis to assess the impact of community-based adherence support on treatment outcomes. Key measures of interest of the ITS were the differences in step change (β) and post-intervention trend (β). RESULTS Short-term, inter-province migrants experienced lower treatment success (aRR = 0.95 [95% CI: 0.92-0.99], p = 0.010) and higher loss to follow-up (aOR = 1.98 [95% CI: 1.44-2.72], p  55 years of age (aRR = 0.93 [95% CI: 0.89-0.96], p < 0.001), relapse patients (aRR = 0.89 [95% CI: 0.84-0.94], p < 0.001), and retreatment patients (aRR = 0.62 [95% CI: 0.52-0.75], p < 0.001) had lower treatment success rates. TB/HIV co-infection was also associated with lower treatment success (aRR = 0.77 [95% CI: 0.73-0.82], p < 0.001) and higher loss to follow-up (aOR = 2.18 [95% CI: 1.55-3.06], p < 0.001). The provision of treatment adherence support increased treatment success (IRR(β) = 1.07 [95% CI: 1.00, 1.15], p = 0.041) and reduced loss to follow-up (IRR(β) = 0.17 [95% CI: 0.04, 0.69], p = 0.013) in the intervention districts. Loss to follow-up continued to decline throughout the post-implementation period (IRR(β) = 0.90 [95% CI: 0.83, 0.98], p = 0.019). CONCLUSIONS Economic migrants, particularly those crossing provincial borders, have higher risk of poor treatment outcomes and should be prioritized for tailored adherence support. In light of accelerating urbanization in many regions of Asia, implementation trials are needed to inform evidence-based design of strategies for this vulnerable population

    Are formalised implementation activities associated with aspects of quality of care in general practice?:A cross-sectional study

    Background: There is a substantial variation in how different general practices manage knowledge implementation, including the degree to which activities are collectively and formally organised. Yet, it is unclear how these differences in implementation activities affect quality of care. Aim: To investigate if there are associations between specific formalised knowledge implementation activities and quality of care in general practices, exemplified by the use of spirometry testing. Design & setting: A nationwide cross-sectional study combining survey and register data in Denmark. Method: An electronic questionnaire was distributed to GPs, and data on spirometry testing among first-time users of medication against obstructive lung diseases were obtained from national registers. Associations were investigated using multilevel mixed-effect logit models. Results: GPs from 1114 practices (58%) responded, and 33 788 patients were linked to a responding practice. In partnership practices, the frequency of interdisciplinary and GP meetings affected the quality of care. Interdisciplinary and GP meetings held on a weekly basis were significantly associated with a higher level of quality of care and this was measured by the odds ratio (OR) of patients having spirometry. The development of practice protocols and standard recordings in the electronic medical record (EMR) for a range of disease areas compared with few or no areas at all also impacted the quality of care level provided. The effect of formalised implementation activities was not as evident in single-handed practices as in partnerships. Conclusion: This study provides valuable knowledge for GPs who aim to organise their practice in a way that supports implementation and quality improvement most effectively. Also, results may be useful for managers of implementation strategies and quality improvement initiatives when planning future activities

    A novel isolator-based system promotes viability of human embryos during laboratory processing

    In vitro fertilisation (IVF) and related technologies are arguably the most challenging of all cell culture applications. The starting material is a single cell from which one aims to produce an embryo capable of establishing a pregnancy eventually leading to a live birth. Laboratory processing during IVF treatment requires open manipulations of gametes and embryos, which typically involves exposure to ambient conditions. To reduce the risk of cellular stress, we have developed a totally enclosed system of interlinked isolator-based workstations designed to maintain oocytes and embryos in a physiological environment throughout the IVF process. Comparison of clinical and laboratory data before and after the introduction of the new system revealed that significantly more embryos developed to the blastocyst stage in the enclosed isolator-based system compared with conventional open-fronted laminar flow hoods. Moreover, blastocysts produced in the isolator-based system contained significantly more cells and their development was accelerated. Consistent with this, the introduction of the enclosed system was accompanied by a significant increase in the clinical pregnancy rate and in the proportion of embryos implanting following transfer to the uterus. The data indicate that protection from ambient conditions promotes improved development of human embryos. Importantly, we found that it was entirely feasible to conduct all IVF-related procedures in the isolator-based workstations

    Novel Approach to Cell Surface Discrimination Between KIR2DL1 Subtypes and KIR2DS1 Identifies Hierarchies in NK Repertoire, Education, and Tolerance

    Cumulative activating and inhibitory receptor signaling controls the functional output of individual natural killer (NK) cells. Investigation of how competing signals impact response, however, has been hampered by the lack of available antibodies capable of distinguishing inhibitory and activating receptors with highly homologous ectodomains. Utilizing a novel combination of monoclonal antibodies with specificity for discrete inhibitory KIR2DL1 and activating KIR2DS1 allotypes found among 230 healthy donors, we investigated allele-specific receptor expression and function driven by KIR2DL1 and KIR2DS1 alleles. We found that co-expression of the HLA-C2 ligand diminishes KIR2DL1, but not KIR2DS1, cell surface staining, but does not impact the respective frequencies of KIR2DL1- and KIR2DS1-expressing cells within the NK repertoire. We can distinguish by flow cytometry NK cell populations expressing the most common KIR2DL1-C245 allotypes from those expressing the most common KIR2DL1-R245 allotypes, and we show that the informative differential binding anti-KIR2DL1/S1 clone 1127B is determined by amino acid residue T154. Although both KIR2DL1-C245 and KIR2DL1-R245 subtypes can be co-expressed in the same cell, NK cells preferentially express one or the other. Cells expressing KIR2DL1-C245 exhibited a lower KIR2DL1 cell surface density and lower missing-self reactivity in comparison to cells expressing KIR2DL1-R245. We found no difference, however, in sensitivity to inhibition or cell surface stability between the two KIR2DL1 isoforms, and both demonstrated similar expansion among NKG2C+ KIR2DL1+ NK cells in HCMV-seropositive healthy individuals. In addition to cell surface density of KIR2DL1, copy number of cognate HLA-C2 hierarchically impacted the effector capacity of both KIR2DL1+ cells and the tolerization of KIR2DS1+ NK cells. HLA-C2 tolerization of KIR2DS1+ NK cells could be overridden, however, by education via co-expressed self-specific inhibitory receptors, such as the heterodimer CD94/NKG2A. Our results demonstrate that effector function of NK cells expressing KIR2DL1 or KIR2DS1 is highly influenced by genetic variability and is calibrated by co-expression of additional NK receptors and cognate HLA-C2 ligands. These findings define the molecular conditions under which NK cells are activated or inhibited, potentially informing selection of donors for adoptive NK therapies
